Intended use
This Thermophilic Reverse Transcriptase or RNA-dependent DNA polymerase is used for real-time fluorescent RT-pyrophosphorolysis activated polymerization (RT-PAP) amplification with either SYBR Green dye or TaqMan probe.
Characteristics
The reverse transcriptase is a thermophilic type A polymerase (US patent pending) with optimal temperatures at 60-62oC, and can be heat-inactivated at ≥90°C.
Particularly, the reverse transcriptase efficiently catalyzes pyrophosphorolysis to remove dideoxynucleotides or acyclonucleotides from the 3’ end of a primer annealed on the RNA/DNA template.
The RTase efficiently synthesizes a complementary DNA strand, one unit per 20 μL of reaction.
The RTase can detect single digit copies of target RNA molecules.
It is particularly suitable to one-step real-time qualitative and quantitative RT-PAP with SYBR Green or TaqMan probe, singleplex or multiplex.
